RESUMO

The black cutworm Agrotis ipsilon is a serious crop pest. Phoxim, an organophosphorus insecticide, has been widely used to control A. ipsilon. When phoxim is extensively applied, the susceptibility of A. ipsilon to insecticide is reduced. However, the mechanisms of tolerance of A. ipsilon to phoxim remain unclear. Herein, we report that an epsilon class glutathione S-transferase, AiGSTE1, confers phoxim tolerance in A. ipsilon. Exposure to a sublethal concentration (LC50) of phoxim caused oxidative stress and activated the transcription of AiGSTe1 genes in A. ipsilon larvae. Recombinant AiGSTE1 expressed in Escherichia coli could metabolize phoxim. Furthermore, E. coli cells overexpressing AiGSTE1 displayed significant tolerance to oxidative stress. Knockdown of AiGSTe1 by RNA interference significantly increased the mortality of A. ipsilon larvae to phoxim. These results demonstrate that AiGSTE1 confers phoxim tolerance in A. ipsilon by metabolizing the insecticide and preventing phoxim-induced oxidative stress.

Smash-ridge tillage is a novel cultivation technique that significantly influences the quality of arable land and crop yield. In this study, we employed high-throughput 16S rRNA sequencing and Biolog-ECO methods to systematically investigate the impact of smash-ridge tillage on soil microbial community structure and functional diversity. The results demonstrate that both ST30 and ST50 treatments significantly enhance the average plant height, average plant diameter, average fresh root weight, stem fresh weight, and leaf area of tobacco plants, with the ST50 treatment exhibiting superior performance. Furthermore, both ST30 and ST50 treatments exhibit significantly higher soil enzyme activity and microbial community diversity compared to the CK treatment. They also improve the soil microbial utilization of carbon sources. Additionally, the ST50-treated soil samples demonstrate 15 microbial functional pathways that exceed those of the CK and ST30 treatments. In conclusion, the Smash-ridge tillage treatment at a depth of 50 cm yields more favorable results. This study provides a theoretical foundation for enhancing soil quality in Smash-ridge tillage by elucidating the mechanisms through which it impacts soil microbial ecology.

Exploring catalysts with low cost and excellent performance for the hydrogen evolution reaction (HER) is still a significant challenge. In this work, zeolitic imidazolate framework 8 (ZIF-8), hybridized with polyvinylpyrrolidone, was used to prepare Zn, Ni co-loaded N-doped porous carbon (ZnNi/NPC) via a straightforward absorption and pyrolysis process. The as-prepared ZnNi/NPC was used as a catalyst for the HER. This experiment showed that the porous structure and Ni doping have a significant influence on the HER activity of the catalyst. Compared with Zn/NC and Zn/NPC, ZnNi/NPC exhibits superior HER activity with an overpotential of 198 mV and a Tafel slope of 69.2 mV dec-1. ZnNi/NPC also shows excellent physical and chemical stability during the HER process. Considering the lower cost and excellent HER performance of ZnNi/NPC, this work provides an attractive solution to fabricate non-precious materials and offers a possible new strategy to replace Pt-based electrocatalysts for HER.

Recently, Ag/AgCl composites with different structures have been widely studied and used as photocatalysts to degrade dye pollutants, due to their high separation efficiency of electron-hole pairs under visible light irradiation. Herein, we adopted a nucleation, precipitation, growth and photoreduction method to prepare Ag and AgCl co-doped ZIF-8 hybrid photocatalysts and explored the influence of Ag content on their physical and chemical properties. All as-prepared samples were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), Brunauer-Emmett-Teller (BET) measurements, energy dispersive spectroscopy (EDS), UV-vis diffuse reflectance and X-ray photoelectron spectroscopy (XPS). XRD indicated that ZIF-8 and AgCl were formed and some of the AgCl was reduced into Ag0 after 30 min of UV light irradiation. SEM and TEM images verified that Ag/AgCl nanoparticles were inlaid in the body of ZIF-8 and Ag ions could hinder the growth of the ZIF-8 crystal. BET data indicated that Ag/AgCl nanoparticles did not alter the pore size of ZIF-8. The UV-vis diffuse reflectance spectra showed that Ag/AgCl@ZIF-8 has excellent ability to absorb visible light, indicating the high efficiency of the electron-hole pair separation of Ag/AgCl@ZIF-8. Finally, the photocatalytic activities of all of the as-synthesized samples were evaluated by degradation of RhB under visible light irradiation. Ag and AgCl co-doped ZIF-8 hybrid photocatalysts exhibited high photocatalytic activity due to the synergistic effect of ZIF-8, AgCl and Ag. After 60 min of visible light irradiation, Ag/AgCl(15)@ZIF-8 exhibited the best photocatalytic activity and could degrade 99.12% RhB, which was higher than Ag/AgCl (94.24%) and ZIF-8 (5.17%). Additionally, a photocatalytic mechanism for dye pollutant degradation over the Ag and AgCl co-doped ZIF-8 hybrid photocatalysts was proposed.

Neuroblastoma is the most common extra-cranial solid tumor in infants and children and accounts for about 15% of deaths from childhood cancers. MicroRNAs (miRNAs) have been shown to play an important role in several cellular processes, such as cell proliferation, apoptosis, invasion, metastasis and angiogenesis, and therefore have been implicated in cancer progression. miR-483-3p is associated with neuroblastoma and is found to function as an 'onco-miR' in some malignancies. However, its role in neuroblastoma remains poorly understood. In this study, we confirmed that miR-483-3p is overexpressed in neuroblastoma tissue when compared with normal tissue and miR-483-3p expression is also associated with tumor stage. Overexpression of miR-483-3p substantially enhanced cell proliferation, migration, and invasion of neuroblastoma cells. miR-483-3p also promoted tumor growth of neuroblastoma in vivo. Both in vivo and in vitro experiments showed that the tumor suppressor PUMA was a target of miR-483-3p. Furthermore, down-regulation of PUMA by small interfering RNA (siRNA) exhibited similar effects to those observed as a result of overexpression of miR-483-3p. Our results indicate that miR-483-3p could function as an 'onco-miR' in human neuroblastoma and reveal a new and potentially important target for neuroblastoma anticancer therapy.

BACKGROUND AND AIM: Increasing evidence indicates that chronic inflammation is a direct or indirect manifestation of hypertension. Potassium channels are thought to be critical for lymphocyte activation, which suggests that hypertension may be an inflammatory disease initiated at the ion channel level. METHODS: This study investigated changes in interleukin (IL)-6, IL-17, and transforming growth factor beta (TGF-b1) expression in the blood of Kazakh hypertensive patients in Northwest China using ELISA technology. Whole-cell patch clamp technology was used to evaluate current changes associated with Kv1.3 and KCa3.1 in peripheral blood T lymphocytes of hypertensive patients, and to investigate current changes induced by telmisartan. We also investigated the effects of telmisartan on expression of Kv1.3 and KCa3.1 at mRNA and protein levels in peripheral blood T lymphocytes using real-time polymerase chain reaction and Western blot analysis. RESULTS: Expression of IL-6, IL-17 and TGF-b1 in the blood of Kazakh hypertensive patients in Northwest China was significantly higher than in healthy controls (p < 0.05). The current mediated by Kv1.3 and KCa3.1 and the corresponding expression at mRNA and protein levels in T lymphocytes were also higher in these hypertensive patients than in controls (p < 0.05). Telmisartan intervention for 24 h and 48 h inhibited the current and expression of Kv1.3 and KCa3.1 at mRNA and protein levels (p < 0.05). CONCLUSIONS: These results indicated that the increase in functional Kv1.3 and KCa3.1 channels expressed in T lymphocytes of Kazakh patients with hypertension was blocked by telmisartan, resulting in a reduced inflammatory response. These results provide theoretical support for the treatment of hypertension at the cellular ion channel level.
